Abstract:

previous studies have shown that morphine addiction is caused by abnormality in neurotransmission system.serotonin is one of the most important neurotransmiters involved in addiction.the dorsal and median raphe nuclei are the important sites that synthesize and release this neurotransmiters in this study we have injected 5,7  dihyroxytryptamine a serotonergic neurotoxin into the dorsal and median raphe nuclei followed of evaluation of craving behaviour for morphine.behavioural and electrophysiological studies(EEG) were conducted for the evaluation of morphine dependence.the results evaluated by self-administration method showed that,5,7 dihyroxytryptamine increased craving for morphine in comparison with sham group.furthermore the results of withdrawal syndrome syndrome sihns showed that 5,7 dihyroxytryptamine increased dependency to morphine compared to the sham group.electrophysiological study revealed that total power of EEG in 5,7 dihyroxytryptamine receiving group was less than the control group(p<0.05) by conclusion it seems that 5,7 dihyroxytryptamine can increase craving to morphine,by destroying serotonergic system.therefore in patients who must receive morphine as an analgesic, prescription of drugs that interact with serotonergic system should be avoided.

Functional mapping of the cardiorespiratory effects of dorsal and median raphe nuclei in the rat.

The dorsal (DRN) and median (MRN) raphe nuclei are important sources of serotonergic innervation to the forebrain, projecting to sites involved in cardiovascular regulation. These nuclei have been mapped using electrical stimulation, which has the limitation of stimulating fibers of passage. The present study maps these areas with chemical stimulation, investigating their influence on cardiores...

Projections of Dorsal and Median Raphe Nuclei to Dorsal and Ventral Striatum

The ascending serotonergic projections are derived mainly from mesencephalic raphe nuclei. Topographical projections from mesencephalic raphe nuclei to the striatum were examined in the rat by the retrograde transport technique of HRP (horseradish peroxidase). In 29 rats stereotaxically injection of HRP enzyme were performed in dorsal and ventral parts of striatum separately. The extent of the ...

effect of diabetes on median and dorsal raphe nuclei efferent fibers to ca3 hippocampal area in rat

diabetes mellitus (dm) is one of the most important metabolic disease causes in many side effects. the decrease of mental abilities and even some form of dementia followed prolonged diabetes has been reported. there are evidences that show hippocampus might be the site of neural involvement in patients with dementia followed diabetes. as hippocampus received main serotonergic efferents from bra...

Cell numbers in the dorsal and median raphe nuclei of AS and AS/AGU rats

The AS/AGU rat is a spontaneous recessive mutation derived from a closed colony of Albino Swiss (AS) rats. This mutation generates a stop codon within the coding region of the PKC-gamma gene. The rats exhibit locomotor dysfunction, which is progressive with age. The nigrostriatal system has already been shown to be affected by this mutation that a) there is marked (80-90%) reduction in extracel...

A Whole-Brain Atlas of Inputs to Serotonergic Neurons of the Dorsal and Median Raphe Nuclei

The serotonin system is proposed to regulate physiology and behavior and to underlie mood disorders; nevertheless, the circuitry controlling serotonergic neurons remains uncharacterized. We therefore generated a comprehensive whole-brain atlas defining the monosynaptic inputs onto forebrain-projecting serotonergic neurons of dorsal versus median raphe based on a genetically restricted transsyna...
